The Clever Boys were having a brilliant run in the league before the second international break, after clinching their fourth consecutive victory by edging Chippa United 1-0.
Gavin Hunt's charges have won this competition in 2017 after narrowly beating Bloemfontein Celtic 1-0 in the final in Durban.
The sides met last season in the same tournament, with Wits knocking out the Team of Choice 2-1 in the quarter-finals but their road was ended in the semi-finals by eventual champions Baroka FC.
Meanwhile, United turned the corner in the Absa Premiership by winning back-to-games against Chippa United and Bakgaga for the first time this season.
Maritzburg have not won the League Cup before but their mentor Eric Tinkler lifted the trophy in 2016 at his former club Cape Town City.
The last time the sides clashed was in February this year in the league, where the Clever Boys edged the Durban-based side 1-0 thanks to a goal by Lehlohonolo Majoro in Johannesburg.
