The Spaniard rubbished talks of Los Blancos being a weaker side than Barcelona, highlighting the need for the Catalan club to be on top of their game.
"Saying Real Madrid are weaker would be foolish, and that's not what we think," he told Marca.
"They have so many qualities that can hurt us, and with the match being at their ground, it's even more important for every aspect of our game to be spot on."
Iniesta also gave an update on Messi, who has been sidelined for about two months due to a knee injury.
"If he's in the squad, it'll be because he's ready, whether it's from the start or on the bench," he added.
"If Leo starts the game, it will improve the team's chances."
Iniesta has played against Real 31 times in his career, winning 13 of those matches and losing 10 times.
