The Gunners made two changes from their win over Brentford, as Oleksandr Zinchenko and Martin Odegaard were recalled. Spurs, meanwhile, were boosted by the return of Son Heung-Min alongside recalls for Cristian Romero and Emerson.
The opening exchanges had plenty of goal-mouth action as Gabriel Martinelli came close to giving Arsenal an early lead with a deflected shot from a half-volley that came off the near post.
After some good chances from either end, the Gunners eventually drew first blood on 20 minutes after Thomas Partey curled a first time shot from the edge of the bix into the top corner from an offload by Ben White.
It took ten minutes for Antonio Conte's charges to restore parity, as Harry Kane expertly dispatched a penalty down the middle after Gabriel brought down Richarlison in the box.
The game continued to open up in the latter phase of the first stanza and Arsenal came the closest to going ahead again just before the half-time break after goalkeeper Hugo Lloris smothered a toe-poked effort by Gabriel Jesus in the box.
Mikel Arteta's men settled the quickest from the break, as Jesus restored their lead within four minutes with a tap-in from a loose ball after a mix-up between Romero and Lloris near the goal-mouth.
Arsenal remained on the front foot as the Lilywhites were reduced to ten men just after the hour-mark after Emerson was red-carded for a lunging tackle on Martinelli, who was going through on goal.
The Gunners capitalised on their numerical advantage five minutes later as Granit Xhaka gave them a two-goal cushion with a low strike under Lloris after Martinelli cut inside the box and played a decisive cut-back.
Conte looked for a response with a quadruple change in the last 20 minutes as Davinson Sanchez, Yves Bissouma, Oliver Skipp and Ryan Sessegnon entered the fray.
However, Arteta's troops did not take their foot off the gas, as they squeezed Spurs in their own half and threatened to run riot. Kieran Tierney came close to making it 4-1 for the Gunners in stoppage time with a low drive towards the bottom corner that was pushed away by Lloris.
The win extended Arsenal's lead at the top of the table to four points while Spurs lost for the first time.
