Nicolas Jackson and Cole Palmer returned to the Blues' starting line-up, while Spurs recalled Micky van de Ven, James Maddison and club captain Son Heung-Min.
Meetings between the two sides have not disappointed in previous, although the Blues have won the last three, including a seven-goal thriller in North London earlier this season.
The opening stages of the first half belonged to the home side, who hit the upright through Jackson's speculative effort while the Senegalese also had an effort saved on the line by Spurs goalkeeper Vicario.
That was the trend for the remainder of the first half as Spurs failed to carve out any clear-cut openings, while Guglielmo Vicario produced a superb save just before the break to deny Jadon Sancho at the other end.
Chelsea got the breakthrough five minutes into the second period when Palmer picked out the run of Enzo Fernandez, who powered home a header from six yards out for his fifth league goal of the season.
Spurs had the ball in the back of the net in the 69th minute when Pape Matar Sarr dispossessed Moises Caicedo before firing home from 20 yards out however after a VAR check, the goal was disallowed for a foul in the build up.
Goalkeeper Robert Sanchez ensured Chelsea walked away with the spoils after pulling off a great save late in the game to deny Son a certain goal, as they secured maximum points.
