Polokwane, on the other hand, qualified for the competition for the first time since 2019 after finishing in eighth spot in the standings last season.
Rise and Shine reached the semi-finals in 2019 but they were knocked out on penalties by Highlands Park after a goalless draw on aggregate.
The last time the sides met in a cup tournament was in the 2021 Nedbank Cup, where Sundowns thrashed City 4-0 in the round of 16 while Rise and Shine were still in the Motsepe Foundation Championship.
Peter Shalulile, Lesedi Kapinga, Lebohang Maboe and Andile Jali were on target for the Tshwane giants.
