The Citizens qualified for Africa's premier competition for the first time in their history after securing a second-place finish in the 2021/2022 DStv Premiership campaign.
City have been involved in continental duties before, as they reached the first round of CAF Confederation Cup under Benni McCarthy but were knocked out by Mozambique's Costa do Sol on the away goals rule.
The Mother City side were in action on Tuesday evening as they played to a disappointing goalless draw against Sekhukhune United in the DStv Premiership, extending their unbeaten run to five matches in the league.
Otoho d'Oyo, meanwhile, have been not been in competitive action since the Congo Premier League concluded in June.
This will also be the Congolese side's second time facing South African opposition, as they lost 5-2 on aggregate to Mamelodi Sundowns in 2019.
