SuperSport dropped points for the second time in their last three matches after they were held to a goalless draw by Marumo Gallants on Tuesday.
United, as a result, were leapfrogged by Orlando Pirates in second spot on the table by two points and could reclaim that position with a win but they would have played a game more.
Matsatsantsa will be without Grant Margeman due to suspension after he picked his fourth booking against Gallants.
AmaZulu, on the other hand, are bidding to end their season in the top eight but they are winless in the last three matches after they came back from two goals down to salvage a 2-2 draw against Cape Town City two weeks ago.
Usuthu are placed 12th on the table and their immediate task would be to get away from the danger of relegation, as they are just three points above the drop zone.
The KwaZulu-Natal side got the better of SuperSport in the first round clash after a come-from-behind 2-1 win.
Thapelo Maseko broke the deadlock for Matsatsantsa but Gabadinho Mhango levelled matters for Usuthu before Bonginkosi Ntuli snatched a late winner.
