Hugo Broos' men bagged their second win in Group C in the last international break after defeating Zimbabwe 3-1 in Bloemfontein.
Bafana will now be taking their clash to the Peter Mokaba Stadium for this evening's clash.
South Africa currently sit third in Group C but they can move top with a victory and hope Nigeria deny Rwanda three points.
Lesotho, meanwhile, will be looking for a return to winning ways after losing 2-0 to Rwanda in their last qualifier in June last year.
The two nations last met in an international friendly last year with the clash ended in a goalless draw in Pretoria.
