Usuthu will be looking to bounce back in the penultimate group B match after their two-game winning streak came to an end following a 2-0 defeat to ES Setif last weekend.
Benni McCarthy's charges dropped down to third on the log, equal on six points with Setif, who edged ahead of them with a better head-to-head scoring record.
The group was pulled wide open after leaders Raja suffered their first defeat after a shock 2-1 loss to Horoya in Guinea last weekend.
The Green Eagles had won three straight matches in the section prior to that, including a 1-0 win over AmaZulu in their group opener, as they remained three points clear at the top.
Raja, as a result, need at least a draw to qualify for the quarter-finals with a match to spare while Usuthu need a win to keep their hopes of qualifying alive.
AmaZulu could be out of contention if they lose and ES Setif beat Horoya in a later game on the same evening.
Usuthu will boosted by the return of fans to the stadium after the club announced that limited tickets were made available on a first-come, first served basis, with all Covid-19 protocols followed.
