Usuthu held their own in their Group B opener against CAF Confederation Cup holders Raja Casablanca away last weekend but suffered a late and narrow 1-0 loss.
Benni McCarthy's charges, as a result, lost their third game in a row in all competitions, after back-to-back defeats to Orlando Pirates in the league and Nedbank Cup.
Similarly, Horoya started their group campaign on a bad note after a narrow 1-0 loss to Algerian side ES Setif.
However, the Guinean outfit are on a nine-game winning run in the domestic league and will be fired up to get off the mark in the section against tournament debutants.
Horoya were in the same group as Kaizer Chiefs in this competition last season but lost out on a quarter-final place to Amakhosi, who edged them on a head-to-head rule to grab second spot after both sides finished equal on nine points.
