The Gunners coach Mikel Arteta made two changes from the big win over PSV Eindhoven as Fabio Vieria and Eddie Nketiah came in for Kai Havertz and Leandro Trossard.
Spurs goalkeeper Guglielmo Vicario was the busier of the two goalkeepers in the opening 15 minutes, as he produced a couple good saves to keep out Gabriel Jesus and Nketiah.
Arsenal took a deserved lead in the 26th minute when Bukayo Saka carved out some space in the box and unleashed a shot that took a big deflection off Christian Romero and sailed into the bottom corner.
Spurs nearly pulled themselves level when Son Heung-min broke free down the right before pulling the ball back for Brennan Johnson, who saw his first-time strike saved by goalkeeper David Raya.
The visitors got their reward shortly before the break as James Maddison got to the byline after skipping past Saka, before pulling the back for Son to tuck home in the six yard area.
Seven minutes into the second half, the Gunners were awarded a penalty after Romero was deemed to have handled the ball in the box by VAR. Saka stepped up and calmly slotted home to restore Arsenal's lead.
The joy did not last long as Maddison won possession off Jorginho and drove towards the box before playing in Son, who calmly slotted past Raya to complete his brace.
With time running out, Arteta withdrew Jesus for Reiss Nelson on 77 minutes while Spurs introduced Richarlison and Pierre-Emile Hojbjerg for Son and Maddison a few moments later.
Both sides were unable to find another clear-cut chance in the second half and were forced to settle for a share of the spoils.
