New signing Donny van de Beek was handed his first start alongside the returning Nemanja Matic and Fred, while Dean Henderson was given the nod between the sticks.
United nearly found the opening goal after 15 minutes when Jesse Lingard's cross fell to Juan Mata, who saw his first-time effort saved by Luton shot-stopper James Shea.
It looked to be another frustrating evening for Ole Gunnar Solskjaer's men as they struggled to carve out clear-cut chances as Ighalo cut a lone figure upfront.
United's efforts were eventually rewarded on the stroke of halftime when Mata converted from the penalty spot after Brandon Williams was brought down in the box.
The visitors went close to pulling two goals ahead on the hour mark when Mata turned provider to pick out Lingard, who saw his strike cleared off the line.
Solskjaer surprisingly opted for a triple change for the final 13 minutes as Marcus Rashford, Mason Greenwood and Bruno Fernandes were introduced.
Henderson was finally called into action on his United debut on 81 minutes, with the shot-stopper showing great reflexes to keep out a towering header at the far post.
The win was sealed with two minutes left on the clock as Fernandes and Greenwood combined to set up Rashford, who calmly slotted into the bottom corner.
Greenwood also managed scoresheet in time added on with a neat finish into the far corner from close range as the Red Devils advance in the League Cup.
