Chiefs could not build momentum to make a case for their league title ambitions after a 1-0 loss to Sekhukhune United last Saturday ended their two-game winning run.
Amakhosi find themselves in fourth place on the standings, three points behind second-placed SuperSport United but they will need to win by at least five goals to topple Matsatsantsa on the log.
AmaZulu, on the other hand, lost for the first time in the league under Romain Folz after going down 2-0 against Royal AM last weekend.
Usuthu currently occupy 12th spot on the table with 18 points, just one point behind eighth-placed Orlando Pirates.
The sides will be meeting for the fourth time this season, with all the three previous clashes ending in draws, including two-legged MTN8 semi-final encounters.
