Jurgen Klopp made just once change from the draw with Chelsea, as James Milner made way for Trent Alexander-Arnold, while Brighton were without Moises Caicedo amid the links with Arsenal.
The Reds went close to breaking the deadlock on four minutes but Mohamed Salah saw his goal-bound strike cleared off the line by Lewis Dunk.
Liverpool had a goal-line clearance of their own in the 15th minute as Alexander-Arnold was in the right place at the right time to keep out Evan Ferguson's effort.
Just minutes after Salah passed up a good chance, the Egyptian turned provider to play in Harvey Elliot who fired into the far corner for his second goal in as many cup games.
Brighton managed to get back on level terms five minutes before the break when Tariq Lampety's shot deflected off Dunk and wrong-footed Alisson Becker.
Both sides came out firing in the second half, with Salah and Thiago Alcantara going close for the away side before the hour mark.
Klopp had seen enough with 25 minutes to go and threw on Milner, Darwin Nunez and Jordan Henderson for Alexander-Arnold, Elliot and Naby Keita.
Both sides pushed for a winner in the latter stages of the second half, but neither were able to get joy in the final third.
That proved to be the trend for the remainder of the half until Brighton broke Liverpool hearts in the the fifth minute of time added on thanks to a fine strike from Kaoru Mitoma that left Alisson with no chance.
