Coach Ruben Amorim made just two changes from the loss to Bournemouth over the weekend, as Leny Yoro and Rasmus Hojlund came in for Tyrell Malacia and Joshua Zirkzee.
The home side looked lively in the early stages of the contest, with United-link Matheus Cunha getting a lot of joy on the ball however the final pass was lacking from the rest of his teammates.
Wolves went close to breaking the deadlock after 25 minutes when Nelson Semedo picked out Jørgen Strand Larsen, who rose above his marker to meet the cross but goalkeeper Andre Onana did well to tip the effort over the crossbar.
The Red Devils, meanwhile, failed to create any openings in the opposition box and they were restricted to efforts from distance, with full-back Diogo Dalot going closest with an effort from 20 yards that was parried away by Jose Sa.
United got off to a poor start in the second half, as captain Bruno Fernandes was sent off a minute in after picking up his second yellow card of the encounter, leaving his side to finish the game with ten men.
The visitors got the breakthrough in the 58th minute when Cunha's corner evaded everybody and sailed directly into the back of the net, with Onana unable to get a hand on the ball.
Red Devils coach Amorim responded with a triple change for the final 30 minutes of the second half as Antony, Casemiro and Christian Eriksen replaced Amad Diallo, Manuel Ugarte and Yoro.
The home side would go on to bag a second goal in second-half stoppage time as a swift counter attack ended with Cunha unselfishly teeing up Hee-Chan Hwang for the simple tap-in for his first goal of the season.
