There was a reunion for former FC Barcelona teammates Lionel Messi and Luis Suarez, although Angel di Maria and Sergio Aguero were named on the bench for a second straight game.
The Argentine's got the early goal in the first half, with Messi turning provider to set up Rodriguez to power a home a header at the far post.
Uruguay, meanwhile, struggled to carve out any openings in the remaining stages of the first half as Suarez and Edinson Cavani cut frustrated figures upfront.
That changed early in the second half as De La Cruz got on the end of Federico Valverde's cross but was unable to find a way past Emiliano Martinez.
Unfortunately, the rest of the stanza failed to provide any clear-cut openings as Lionel Scaloni's charges got their first win at the Copa America.
"I honestly believe that in the previous matches we also deserved to win, but due to small mistakes we were only able to draw," Scaloni said after the game, as per Stadium Astro.
"Today the team gave the extra rage so that they don't convert us and once we were winning we were able to withstand the opponent's attacks. Today we were good both in attack and defence."
