However, the Spaniard didn't want to reveal whether Suarez would start immediately and how long he will play.
"He will have some minutes for sure, how many I cannot say," Enrique said in his pre-match press conference.
"It is a big thing for him, to join up finally with the team and get some minutes."
Enrique, who played five years at Madrid, but later became a club hero at Barcelona during his playing days, insisted he cannot foresee how Suarez would cope with the pressure of debuting in such a massive game.
"Each case is totally different," he continued.
"It depends a lot on the personality of the player."
The Barcelona boss, who will be on the side-lines in an El Clasico for the first time, has revealed he will set up his side to control the game.
"We will be coming out against Madrid with the same attitude we have shown all season," the 44-year-old added.
"We'll go for the game from the start, look to have control of the ball in their half. We will see if we're capable of that or not."
