Omar Marmoush, Ilkay Gundogan and Rico Lewis were recalled to the City line-up, as Phil Foden, Bernardo Silva and Matheus Nunes all dropped to the bench.
The hosts got off to the perfect start after being awarded a penalty as Marmoush was brought down in the box by Webster, which allowed Erling Haaland to step up and calmly slot home on 10 minutes.
In doing so, Haaland became the first player in Premier League history to reach 100 goal involvements in fewer than 100 appearances, having done it 94.
Brighton did get back on level term in the 21st minute as Pervis Estupiñán opened his account for the season with a delightful free-kick that gave Stefan Ortega with no chance.
However, Pep Guardiola's charges restored their lead minutes before the halftime break as Marmoush collected a pass from Gundogan before unleashing a powerful strike from 20 yards that sailed into the back of the net.
Three minutes into the second half, Brighton were back on level terms as a goal-mouth scramble in the City box ended with defender Abdukodir Khusanov deflecting the ball into his own net.
Guardiola introduced Silva just after the hour mark for Savinho, but he only lasted just under 20 minutes as he was forced off with an injury and replaced by Foden for the final 15 minutes of the second half.
City almost bagged the third goal with 10 minutes left on the clock when Nico Gonzalez rose highest to meet a corner from Gundogan, but his header crashed against the upright as the encounter ended all square.
